AtDAGT2 is a functional CoA:diacylglycerol acyltransferase with preference for polyunsaturated fatty acids
Abstract
Demonstration of the function of the Arabidopsis DGAT2 (AtDGAT2) has remained elusive despite biochemical testing of genetic mutants and overexpression lines.This is in contrast to several other plant species such as castor and tung where the activity of DGAT2 has been biochemically confirmed. We have expressed AtDGAT2 in the Nicotiana benthamiana transient leaf assay and demonstrated acyl-CoA:diacylglycerol acyltransferase activity. Transient expression of AtDGAT2 in N. benthamiana leaves resulted in an increase in triacylglycerol (TAG) twice as great as the increase observed following parallel expression of AtDGAT1. Similarly, biochemical analysis showed higher conversion rate from labeled diacylglycerol (DAG) to TAG in the case of AtDGAT2 compared to AtDGAT1. In addition, AtDGAT2 had a higher preference for linoleoyl-CoA and linolenoyl-CoA than oleoyl-CoA as the acyl donor.These results allow us to conclude that AtDAGT2 is a functional acyl-CoA:diacylglycerol acyltransferase enzyme that can catalyze substantial increase in TAG synthesis.
